OBJECTIVES/GOALS: The Institute of Translational Health Science (ITHS) Remote Technologies for Research Reference Center (REMOTECH) aims to support researchers using remote technologies. Understanding barriers specifically for engagement of diverse populations is critical to improve equitable access and increase diverse participation in research. METHODS/STUDY POPULATION: We conducted semi-structured interviews with researchers (N=30) within the WWAMI (Washington, Wyoming, Alaska, Montana and Idaho) region, identified through participation in a previous survey as well as those known to have previously implemented remote technologies at ITHS institutions. We solicited specific concerns regarding equity, diversity, and inclusion (EDI) related to remote research participant recruitment, retention, and implementation, as well as outcomes and potential solutions. Interview transcripts were coded, summarized, and emerging themes were identified. RESULTS/ANTICIPATED RESULTS: The pandemic necessitated a shift to the use of remote research engagement strategies. Our interview findings show that researchers have a desire to: increase diversity through remote engagement options; decrease participant burden; and understand and include strategies that advance equity, diversity, and inclusion (EDI) efforts. While geographic diversity was positively impacted by the use of remote technologies, significant barriers and challenges currently exist in the use of remote technologies with respect to other types of diversity, for example access to technologies and limited financial resources. DISCUSSION/SIGNIFICANCE: Remote technologies in research could increase diversity, but interviewees shared barriers and challenges that prevent that from happening. Researchers would benefit from equity-oriented digital literacy resources to use with diverse populations. Such tools can also be used to guide study design/procedures, materials, and processes.

Following the emergence of SARS-CoV-2, early outbreak response relied on behavioural interventions. In the USA, local governments implemented restrictions aimed at reducing movements and contacts to limit viral transmission. In Pennsylvania, restrictions closed schools and businesses in the spring of 2020 and interventions eased later through the summer. Here we use passive monitoring of vehicular traffic volume and mobile device-derived visits to points of interest as proxies for movements and contacts in a rural Pennsylvania county. Rural areas have limited health care resources, which magnifies the importance of disease prevention. These data show the lowest levels of movement occurred during the strictest phase of restrictions, indicating high levels of compliance with behavioural intervention. We find that increases in movement correlated with increases in reported SARS-CoV-2 cases 9–18 days later. The methodology used in this study can be adapted to inform outbreak management strategies for other locations and future outbreaks that use behavioural interventions to reduce pathogen transmission.
Postemergence Control and Glyphosate Tolerance of Doveweed (Murdannia nudiflora)

Doveweed is a problematic weed of lawns and sod production, as well as golf course roughs, fairways, and tees. End-user reports of selective POST control options are inconsistent and control is often short-lived. In addition, inconsistent control with non-selective herbicides such as glyphosate is common. The goals of this research were: (1) evaluate selective POST doveweed control options in ‘Tifway’ hybrid bermudagrass turf; (2) compare efficacy of single vs. sequential applications of selective POST herbicides; (3) quantify doveweed tolerance to glyphosate; and (4) quantify recovery of foliar applied glyphosate following treatment with a C14-glyphosate solution. A single application of sulfentrazone+metsulfuron; thiencarbazone+iodosulfuron+dicamba or 2,4-D+MCPP+dicamba+carfentrazone; or thiencarbazone+foramsulfuron+halosulfuron provided >60% control 2 weeks after initial treatment (WAIT). A second application of these treatments 3 WAIT improved control 6 WAIT. Two applications of 2,4-D+MCPP+dicamba+carfentrazone or thiencarbazone+foramsulfuron+halosulfuron provided ~80% control 6 WAIT. Doveweed was tolerant to glyphosate application up to 5.68 kgaeha-1. Absorption of 14C-glyphosate was compared between doveweed with cuticle intact, doveweed with a disturbed cuticle, and smooth crabgrass. 14C-glyphosate recovery from the leaf surface of doveweed plants with an intact cuticle was 93.6%. In comparison, 14C-glyphosate recovery from the leaf surface of doveweed plants with a disrupted cuticle and the leaf surface of crabgrass plants was 79.1 and 70.5%, respectively.

Background
A few studies have evaluated the impact of clinical trial results on practice in paediatric cardiology. The Infant Single Ventricle (ISV) Trial results published in 2010 did not support routine use of the angiotensin-converting enzyme inhibitor enalapril in infants with single-ventricle physiology. We sought to assess the influence of these findings on clinical practice.
MethodsA web-based survey was distributed via e-mail to over 2000 paediatric cardiologists, intensivists, cardiothoracic surgeons, and cardiac advance practice nurses during three distribution periods. The results were analysed using McNemar’s test for paired data and Fisher’s exact test.
ResultsThe response rate was 31.5% (69% cardiologists and 65% with >10 years of experience). Among respondents familiar with trial results, 74% reported current practice consistent with trial findings versus 48% before trial publication (p<0.001); 19% used angiotensin-converting enzyme inhibitor in this population “almost always” versus 36% in the past (p<0.001), and 72% reported a change in management or improved confidence in treatment decisions involving this therapy based on the trial results. Respondents familiar with trial results (78%) were marginally more likely to practise consistent with the trial results than those unfamiliar (74 versus 67%, p=0.16). Among all respondents, 28% reported less frequent use of angiotensin-converting enzyme inhibitor over the last 3 years.
ConclusionsWithin 5 years of publication, the majority of respondents was familiar with the Infant Single Ventricle Trial results and reported less frequent use of angiotensin-converting enzyme inhibitor in single-ventricle infants; however, 28% reported not adjusting their clinical decisions based on the trial’s findings.
A Pro106 to Ala Substitution is Associated with Resistance to Glyphosate in Annual Bluegrass (Poa annua)

Glyphosate is used in the transition zone to control annual bluegrass in fully dormant warm-season grasses. A suspected resistant (R) biotype of annual bluegrass was identified on a golf course in South Carolina after at least 10 consecutive years of glyphosate application. Greenhouse bioassays revealed the R biotype was 4.4-fold resistant to glyphosate compared with a standard susceptible (S) biotype. Further studies were conducted to investigate the mechanism conferring glyphosate resistance in the R biotype. Leaf discs of both biotypes accumulated shikimate in response to increasing glyphosate concentration, but the glyphosate concentration resulting in 50% EPSP synthase inhibition as a result of shikimate accumulation (I50) was 4.2-fold higher in the R biotype compared with the S biotype. At the whole plant level, similar levels of shikimate accumulation were observed between biotypes at 6 and 24 h after treatment (HAT) with glyphosate, but greater shikimate accumulation occurred in the S biotype at 72, 120, and 168 HAT. Shikimate levels decreased in the R biotype after 72 HAT. There were no differences in 14C-glyphosate absorption between biotypes. However, more 14C-glyphosate translocated out of the treated leaf in the R biotype and into root tissues over time compared with the S biotype. Partial sequencing of the EPSP synthase gene revealed a point mutation that resulted in an Ala substitution at Pro106. Although other mechanisms may contribute to glyphosate resistance, these results confirm a Pro106 to Ala substitution is associated with resistance to glyphosate in the R annual bluegrass biotype.
Functional health status in children and adolescents after Fontan: comparison of generic and disease-specific assessments

Purpose: The aim of this study was to compare associations between generic versus disease-specific functional health status assessments and patient and clinical characteristics for patients with severe congenital heart disease. Methods: This was a cross-sectional observational study involving 325 single ventricle patients, aged 10–18 years, after Fontan procedure. Enrolled patients underwent a medical history review, laboratory testing, and assessment of the functional health status by completion of the generic Child Report Child Health Questionnaire and the disease-specific Congenital Heart Adolescent and Teenage questionnaire. Correlated conceptually equivalent domains from both questionnaires were identified and their associations with patient and clinical variables were compared. Results: From the generic assessment, patients perceived marginally lower physical functioning (p = 0.05) but greater freedom from bodily pain compared with a normal population (p < 0.001). The equivalent physical functioning/limitations domain of the generic instrument, compared with the disease-specific instrument, had similar associations (higher multi-variable model R2) with medical history variables (R2 = 0.14 versus R2 = 0.12, respectively) and stronger associations with exercise testing variables (R2 = 0.22 versus R2 = 0.06). Similarly, the corresponding freedom from bodily pain/symptoms domains from both questionnaires showed a greater association for the generic instrument with medical history variables (R2 = 0.15 versus R2 = 0.09, respectively) and non-cardiac conditions (R2 = 0.13 versus R2 = 0.06). The associations of each questionnaire with echocardiographic results, cardiac magnetic resonance imaging results, and serum brain natriuretic peptide levels were uniformly weak (R2 range <0.01 to 0.04). Conclusions: Assessment of the physical functional health status using generic and disease-specific instruments yields few differences with regard to associations between conceptually similar domains and patient and clinical characteristics for adolescents after Fontan procedure.
Bone Chemistry at Cerro Oreja: A Stable Isotope Perspective on the Development of a Regional Economy in the Moche Valley, Peru During the Early Intermediate Period

In this paper we test the hypothesis that an intensification of maize production preceded the development of a regional Moche political economy in the Moche Valley of north coastal Peru during the Early Intermediate period (400 B.C.—A.D. 600). To do so we analyze stable isotopic signatures of 48 bone apatite and 17 tooth enamel samples from human remains recovered from the site of Cerro Oreja, a large urban and ceremonial center in the Moche Valley. These remains date to the Guañape, Salinar, or Gallinazo phases and provide a diachronic picture of subsistence before the appearance of the Southern Moche state. The most notable patterns identified in the study include a lack of significant change in δ13 Capatite values from the Guañape to Satinar phases, followed by a significant enrichment in δ13 Capatite values from the Salinar to Gallinazo phases. Several lines of evidence, including archaeological context, dental data, and comparative carbon stable isotope data from experimental animal studies and studies of archaeological human remains support the interpretation that the observed 13C enrichment in stable isotope values in the Gallinazo phase primarily reflects maize intensification. The stable isotope data from Cerro Oreja thus suggest that a shift in subsistence toward a highly productive and storable crop may have served as an important precursor to state development during the Early Intermediate period in the Moche Valley.

The original authors of Billman et al. (2000) are joined by three other analysts from the Cowboy Wash research team to respond to the critique of this article by Dongoske et al. (2000). Dongoske and his coauthors state that Billman et al. (2000) failed to test alternative hypotheses or to consider alternative explanations for the findings at 5MT10010 and similar sites. The original authors point out that alternative hypotheses were examined and rejected, leaving a violent episode of cannibalism as the most plausible explanation for the remains found at 5MT10010. Dongoske et al. also question many aspects of the osteological, archaeological, coprolite, and biochemical analyses that were presented in the 5MT10010 study. Our response addresses issues of data collection, procedure, and interpretation, and attempts to clarify some points that were not fully developed in the original text due to length restrictions.

The existence of cannibalism has emerged as one of the most controversial issues in the archaeology of the American Southwest. In this paper, we examine this issue by presenting the results of our investigation at 5MT10010, a small early Pueblo III habitation site in southwestern Colorado. Battered, broken bones from seven individuals were discovered in two adjacent pithouses at 5MT10010. Mixed and incomplete remains of four adults and an adolescent were recovered from the floor and ventilator shaft of one pithouse; the remains of two subadults were found on the floor and in various subfeatures of the second. Cut marks and percussion scars implicate humans in the disarticulation and reduction of these bodies. Evidence of heat exposure on some bone fragments and laboratory analyses of a human coprolite recovered from one of the pithouses support the interpretation that people prepared and consumed human body parts. The discovery of disarticulated human remains at 5MT10010 is one of a number of similar finds in the northern Southwest. Analysis of cases from the Mesa Verde region indicates a sharp increase in cannibalism around A.D. 1150, a time of drought and the collapse of the Chaco system. The causes, consequences, and nature of this apparent outbreak of cannibalism are examined in light of 5MT10010 and other recent finds.
